白藜芦醇对早期心衰大鼠模型血流动力学的影响 被引量:1

Effects of resveratol on hemodynamics in early heart failure model rats

摘要 目的研究白藜芦醇对早期心衰模型大鼠的作用。方法 SD大鼠40只分为假手术组、模型组和白藜芦醇(10、20和40 mg·kg-1)组(均n=8),除假手术组外,其他组大鼠结扎左冠状动脉前降支制心肌梗死致早期心衰模型,制模期间灌胃给予相应剂量的溶剂或药物,共6周。采用血流动力学和心脏称重检测心功能,测定血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、一氧化氮(NO)和一氧化氮合酶(NOS)含量。结果制模6周后,模型组大鼠左心室收缩末压(LVSP)和左心室等容收缩期室内压最大上升速率(+dp/dtmax)显著下降,左心室舒张末压(LVEDP)和左心室等容舒张期室内压最大下降速率(-dp/dtmax)也显著降低,左心重/全心重均显著下降,SOD显著减少,MDA、NO和NOS显著增加,与假手术组相比有显著差异(均P<0.05);白藜芦醇20、40 mg·kg-1组大鼠LVSP和+dp/dtmax显著上升,白藜芦醇40 mg·kg-1组LVDP和-dp/dtmax显著高于模型组(P<0.05)。白藜芦醇20、40 mg·kg-1组SOD增加,MDA、NO和NOS显著降低,左心重/全心重均高于模型组(P<0.05)。讨论白藜芦醇可改善早期心衰大鼠模型的心功能,抗氧化能力增强。 AIM To study the effects of resveratol used in the treatment of early heart failure in experimental rat model. METHODS Forty SD rats were divided into sham, model and resveratrol groups (10, 20, 40 mg·kg^-1, ig) . Heart failure was indeced with ligature of the left coronary artery in rats. The indicators of cardiac function were test by the ways of hemodynamic. The cardiac weight and serum superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO) and nitric oxide synthases (NOS) were measured in each group. RESULTS Compared with the sham group, left ventricular systolic pressure (LVSP), +dp/dtmax, left ventricular end- diastolic pressure (LVEDP) and -dp/dtmax were significantly decrease, left ventricular weight/heart weight were also decreased, the activity of SOD was lower and the level of MDA, NO and NOSwere increased in the model group (P 〈 0.05) . Furthermore, above indicators could be changed by resveratrol 20 or 40 mg·kg^-1 (P 〈 0.05) . CONCLUSION Resveratol has protective effect on early heart failure in the experimental rat model, in which may be related to improving heart function and enhance the body' s antioxidant capacity.
