摘要
目的:建立虎杖中白藜芦醇及其糖苷异构体含量的测定方法。方法:以甲醇提取目标组分,以聚酰胺薄膜为固定相,苯-甲醇-甲酸(10:5:1)为展开剂层析分离,用荧光扫描法定量,λ_(Ex)=306nm,λ_(Em)=400nm,线性参数SX=3。结果:4种组分(顺式白藜芦醇及其糖苷和反式白藜芦醇及其糖苷)的线性关系良好,相关系数为0.9942~0.9996,平均回收率为97.6%~98.6%,RSD为1.8%~3.1%。测定4个虎杖样品中反式白藜芦醇和反式白藜芦醇苷的含量分别为0.20%~0.22%和2.04%~2.14%,均未检出可定量测定的顺式异构体。结论:方法简便、快速、灵敏、测定成本低。
Objective :To develop a method for determining resveratrol and piceid in Polygonum cuspidatum Sieb. et Zucc. by TLC fluorescence scanning, nethods:Resveratrol and piceid were extracted by methanol. The separation was accomplished on polyamide plate by development using the mobile phase benzene- methanol- formic acid (10: 5: 1). The contents of resvertrol and piceid were determined by TLC fluorescence scanning and λEx = 306 nm, λEm = 400 nm,SX = 3. Results: The calibration curves showed good linearity and the interrelated ratio was 0. 9942 - 0. 9996. The average recoveries were 97. 6% - 98. 6% with RSD1.8% - 3. 1%. 4 samples were determined by the method. The level of trans - resveratrol and trans - piceid were 0. 20% - 0. 22% and 2. 04% - 2. 14%, respectively. Conclusion:The results show that the method is convenient ,rapid ,sensitive and low cost.
