磷酸肌酸钠及丹参酮ⅡA磺酸钠对阿霉素心肌病大鼠心室肌细胞单通道钙电流作用研究 被引量:1

The Effeces of Sodium Phosphocreatine and Sodium Tanshinone Ⅱ-A Sulfonate on Single Channel Ca^2+ Current of Ventricular Myocytes in the Dilated Cardiomyopathy Mice Induced by Adriamycin

摘要 目的观察磷酸肌酸钠及丹参酮磺酸钠酮ⅡA磺酸钠对阿霉素心肌病大鼠心室肌细胞单通道钙电流的影响。方法 40只wister雄性大鼠分为磷酸肌酸钠+阿霉素组干预组(n=10),丹参酮ⅡA磺酸钠+阿霉素干预组(n=10),阿霉素组(n=10)和正常对照组(n=10)。观察大鼠的一般情况,14d处死取心脏标本,Langendorff离体心脏逆向灌流法分离大鼠心室肌细胞,随机选取心室肌细胞分为对照组、模型组、模型+磷酸肌酸钠组和模型+丹参酮ⅡA磺酸钠组。应用细胞贴附式方式记录心室肌细胞钙电流,分析对钙单通道电流的影响。结果磷酸肌酸钠和丹参酮ⅡA磺酸钠可使模型组心室肌细胞钙通道开放时间,开放概率明显降低(p<0.05),关闭时间明显增高(p<0.05)。结论丹参酮ⅡA磺酸钠和磷酸肌酸钠通过降低心室肌细胞钙通道开放时间、增加关闭时间、降低开放概率抑制心室肌细胞钙电流。 Objective To Observe the effeces of sodium phosphocreatine and sodium Tanshinone Ⅱ-A sulfonate on the Single Channel Ca2+ Current of ventricular myocytes in the dilated cardiomyopathy mice induced by Adriamycin. Methods The 40 wister male mice were randomly divided into control group(n=10), adriamycin group(n=10), Creatine phosphate sodium + adriamycin group(n=10) and sodium Tanshinone Ⅱ-A sulfonate+ adriamycin group group(n=10). On 14 days, all the mice were sacrificed to take heart, perfused heart with Langendorff, separated ventricular myocytes with Reverse perfusion. Recorded Ca2+ current of ventricular myocytes with the way of cell-attached, and analyzed the impaction to Single Channel Ca2+ Current. Results Compared with that of adriamycin group, sodium phosphocreatine and sodium Tanshinone Ⅱ-A sulfonate groups reduced opening hours of Ca2+ channel of ventricular myocytes in the dilated cardiomyopathy mice induced by Adriamycin(p 〈0.05); opening probability was significantly lower(p 〈0.05), and the closing time was significantly increased(p 〈0.05). Conclusion The sodium phosphocreatine and the sodium Tanshinone Ⅱ-A sulfonate reduced opening hours,increased closing time, and reduced the opening probability of the Ca2+ channel of the ventricular myocytes in the dilated Cardiomyopathy mice induced by Adriamycin.
