摘要
用20例扩张型心肌病患者血清中抗心肌线粒体ADP/ATP载体抗体进行心肌细胞毒性试验,发现18例患者血清引起心肌细胞毒性损害,且具有钙依赖性,异搏停明显地抑制这种细胞毒性反应,使细胞死亡率均值从71.61%±7.67%降到42.56%±6.54%(P<0.01),该细胞毒性反应与对照组比较具有显著性差异.本研究提示抗心肌线粒体ADP/ATP载体抗体作用于心肌细胞膜钙通道,引起细胞钙超负荷和细胞死亡,是扩张型心肌病的发病机理之一.
The myocardial cytotoxic test was performed using the antibodies against the mito-chondrial ADP/ATP carrier in 20 patients with dilated cardiomyopathy. The sera induced myocardial cytotoxicity in 18 out of the 20 patients. The cytotoxicity was calcium-dependent and was-markedly inhibited by verapamil ,the mean cell mortality(MCM) was reduced from 71. 61%±7. 67 % to 42.56% ±6. 54%(P<0.01), compared with the MCM of control group and had significant difference. The above findings suggest that the antibodies against the ADP/ATP carrier bind to the calcium channel of the myocardial cell membrane, induce intracellular calcium overload and cell death. It may be one of the pathogenesis in dilated cardiomyopathy.
