摘要
目的研究mi R-217靶向MAPK1与胃癌侵袭转移的关系。方法在3个不同分化的胃癌细胞株MKN-28、SGC-7901、BGC-823中通过q RT-PCR技术检测mi R-217情况,Western blot检测MAPK1表达,Transwell实验分析mi R-217下调MAPK1表达后胃癌细胞侵袭转移能力改变。结果低分化人胃癌细胞株中MAPK1低表达,而mi R-217高表达,中、高分化人胃癌细胞株中MAPK1高表达,而mi R-217低表达(P<0.05);SGC-7901-mi R-217mimics细胞株中mi R-217表达增高,而MAPK1表达下调(P<0.05);而SGC-7901、SGC-7901-mi R-Scramble、SGC-7901-mi R-217-PM 3个细胞株中mi R-217表达水平较低,而MAPK1表达水平较高(P>0.05)。SGC-790-mi R-217 mimics侵袭细胞计数明显低于其他3组(P<0.05),SGC-7901、SGC-7901-mi R-Scramble、SGC-7901-mi R-217-PM 3组之间侵袭细胞计数无显著性差异(P>0.05)。结论 mi R-217能靶向结合MAPK1,降低其表达水平并抑制胃癌细胞的侵袭和转移。
Objective To study the relationship between miR-217 targeted to MAPK1 and the metastasis,invasion of gastric cancer cells. Methods The level of miR-217 was detected by qRT-PCR in MKN-28,SGC-7901 and BGC-823 of gastric cancer cell lines. The expression of MAPK1 was detected by Western blot. And the extent of ability change of metastasis and invasion of gastric cancer cells were examined by Transwell. Results Low expression of MAPK1 and high expression of miR-217 appeared in BGC-823. High expression of MAPK1 and low expression of miR-217 appeared in MKN-28 and SGC-7901(P〈0.05). Expression of miR-217 was higher and the expression of MAPK1 was lower in SGC-7901-miR-217 mimics(P〈0.05). Low expression of miR-217 and high expression of MAPK1 appeared in SGC-7901,SGC-7901-miR-Scramble,SGC-7901-miR-217-PM(P〉0.05). The number of invaded cells was smaller in SGC-7901-miR-217 mimics than other groups (P〈0.05). Conclusion miR-217 can be the target for MAPK1 and reduce the expression ability, inhibit the metastasis and inva-sion of gastric cancer cells.
