摘要
目的观察五味子乙素诱导人胃癌细胞株MGC-803凋亡作用。方法用10、50、100、200μmol/L浓度五味子乙素作用胃癌细胞株48 h,倒置显微镜下观察细胞增殖情况;MTT法检测五味子乙素对细胞增殖的抑制作用;电镜观察、判定凋亡细胞;凋亡检测试剂盒检测细胞凋亡。结果倒置显微镜下,随着五味子乙素作用浓度升高,细胞增殖能力随之下降;MTT法亦检测到上述结果;当五味子乙素作用浓度是100μmol/L时,可检测到明显的凋亡细胞。结论高浓度的五味子乙素可剂量依赖性地诱导胃癌细胞株MGC-803细胞的凋亡。
Objective To observe the effects of Schisandrin B on apoptosis of human gastric cancer cell line MGC-803.Methods The gastric cancer cells were treated with 10,50,100 and 200 μmol/L Schisandrin B 48 h,and the cells were observed under inverted microscope.MTT assay was used to evaluate the inhibitory effects of Schisandrin B on the cancer cells.Observation and assessing apoptosis cells were under electron microscope.Apoptosis detective kit was used to detect the apoptosis.Results With the increasing of drug concentration,the proliferation of the cells decreased;The same result was gotten by MTT assay.When the cells were treated with 100 μmol/L Schisandrin B,apoptosis cells were detected.Conclusions High concentration Schisandrin B can induce the apoptosis in gastric cancer cell line MGC-803,which is in concentration-dependent manner.
