摘要
残留在动物体内的盐酸克伦特罗可以在人体内富集并引发一系列严重的毒副现象,因而本研究建立了基于固定化抗体的CL直接竞争ELISA检测方法。结果表明由该方法得出的标准曲线的线性回归方程为y=-0.1258Ln(x)+0.6219(y为结合率B/B0,x为标准CL浓度(ng/m L)),R为0.9784,IC50为2.64ng/m L,其线性检测范围为0.24-28.61ng/m L,检测限为0.0859ng/m L,而且和沙丁胺醇,特布他林,肾上腺素均无明显的交叉反应。同时该方法对样品肉中CL的加标回收率均高于85%,变异系数均小于20%。本研究结果可为采用免疫传感器法测定CL提供理论和应用基础。
Clenbuterol in animal tissue can be enriched in human body, and then result in human food poisoning. In this work, a direct competitive ELISA method based on immobilited antibody was established to detect CL rapidly. The results suggested that the linear equation was y=-0.1258Ln(x)+0.6219(R2=0.9784), ICs0 was 2.64ng/mL, the limits of detection for CL were in the range of 0.24-28.61ng/mL and the limit of detection was 0.0859ng/mL. The cross reaction experiment of other β2-receptor agonists showed that the cross-reactivity with salbutamol, terbu- taline and adrenaline was less than 0.1 percent inhibition rate anti CL. The recoveries of CL from pork samples were all above 85%, the variation coefficient was below 20%. The results of this study may provide a theoretical basis for the determination of CL by immune sensor method.
