摘要
采用响应面法对抗盐酸克伦特罗抗体的固定化条件进行研究。结果表明:当固定温度4℃,固定方式振荡,抗体稀释倍数5.325×105,料液比0.948 mL/颗,固定时间11.372 h时,固定化抗体活力最高,即HRP酶标IgG底物显色反应的OD可达1.497 8。优化后的实验值与模型预测值(1.501 3)基本相符。
The immobilization conditions of the monoclonal antibody anti-clenbuterol were optimized by re- sponse surface methodology. The results showed that the immobilized temperature was 4 ℃, the immobi- lized way was used for oscillation, the stationary phase was pH 9.6 carbonate buffer, antibody dilution ratio was 1:532 500, the ratio of solid to liquid was 0. 948 mL per stars, and the fixed time was 11. 372 h. Under such conditions, the TMB substrate termination reaction OD against enzyme labeled an- tibody could reach 1. 497 8. The obtained value of optimized immobilization was in good agreement with the predicted value of 1. 501 3.
