摘要
目的评估以5-氨基酮戊酸为光敏剂的光动力疗法在体外对红色毛癣菌的抑制作用。方法以96孔细胞培养板为实验载体,加入红色毛癣菌悬液,分为治疗组和4个对照组,其中治疗组同时给予氦氖激光照射、加入5-氨基酮戊酸、共同避光孵育12 h等三个条件;而4个对照组均不红色毛癣菌同时具备治疗组所获得的三个条件。然后按照设定的实验条件进行5-氨基酮戊酸光动力学治疗,并用噻唑蓝法检测红色毛癣菌的存活率。结果 5-氨基酮戊酸的浓度介于0.05%~2.5%之间时,治疗组红色毛癣菌的存活率依次递减,对应的存活率之间进行两两比较,差异均有统计学意义(P〈0.05),存活率对5-氨基酮戊酸的浓度呈剂量依赖关系。而当5-氨基酮戊酸浓度介于2.5%~10%时,治疗组红色毛癣菌的存活率差异无统计学意义(P〉0.05),当5-氨基酮戊酸的浓度介于0.05%~10%之间时,对照组中红色毛癣菌的存活率未受影响,四组的红色毛癣菌的存活率变化无明显差异(P〉0.05)。结论在特定浓度下,5-氨基酮戊酸光动力学疗法对红色毛癣菌有明显的抑制作用。
Objective To evaluate the effect of 5-aminolevulinic acid photodynamic therapy on Trichophyton rubrum in vitro. Methods 96-well cell culture plates were used as experimental carriers for adding Trichophyton rubrum suspension, which were divided into experimental group and four control groups. He-Ne laser irradiation,5-aminolevulinic acid and avoiding light incubation for 12 hours were given in the experimental group at the same time. But four control groups did not received three conditions at the same time. Then ALA-PDT was done under experimental conditions. The survival rate of Trichophyton rubrum was detected by MTT. Results When the concentration of ALA ranged from 0.05% to 2.5%, the survival rate of Trichophyton rubrum decreased in experimental group(P0.05). The survival rate and the concentration of ALA were in a dose-dependent manner. When the concentration of ALA ranged from 2.5% to 10%, there was no significant difference in the survival rate of Trichophyton rubrum(P〉0.05). The survival rate of Trichophyton rubrum in four control groups were not affected, and there was no significant difference(P〉0.05). Conclusion ALA-PDT could effectively inhibit the growth of Trichophyton rubrum when ALA is within specific concentration range.
