摘要
文冠果良种选育时间长,优树数量少,用于组织培养的营养器官外植体更少,能否用少量试材减少消毒损失和在较短时间内获得更多的无菌外植体,成为该树种优树组织培养扩繁的关键环节。本文以文冠果休眠枝条茎段为试材,采取不同消毒方法,MS为基本培养基,进行组织培养,结果表明:文冠果茎段消毒的最佳方式是5%NaClO点滴叶腋处+75%酒精30s+0.2%HgCl28min(附加吐温-20 2滴)+75%酒精30s;诱导茎段产生不定芽的最佳激素组合是MS+6-BA1.0mg/L+IAA2.0mg/L;茎段在最佳激素组合下培养67d平均可切取新芽12.10株。
The time to select Xanthoceras sorbifolia's improved variety of seed is long and the Champion tree's number is poor,so that there are not enough vegetative organs explants are used in doing tissue culture.Whether can we use a small amount of material,reduce the loss in disinfecting and get more bioclean explants in short time is becoming the key link to the tissue culture propagation of the champion tree.This paper selected different disinfection methods and Xanthoceras sorbifolia's dormant branches were cultured with MS medium in order to do tissue culture The results showed that;the optimal way to disinfect is 5%NaClO drip axils +75%alcohol30s +0.2%HgC128min(add 2 drops of Tween-20) +73%alcohol 30s;the optimal hormone combination for stems induced adventitious buds is MS +6- BAl.Omg / L + IAA2.0mg / L;in optimal hormone combination,stem segments cultured in 67 d can get12.10 buds on average.
