文冠果叶抑制人肝癌细胞HepG2增殖和抗氧化活性部位的筛选 被引量:4

摘要 目的:研究文冠果叶不同萃取物的抗氧化及抗肝癌活性。方法:分别用水、30%乙醇水溶液、50%乙醇水溶液、70%乙醇水溶液及95%乙醇水溶液萃取文冠果叶,得到5个不同的提取部位A、B、C、D、E,采用MTT法筛选对人肝癌细胞HhepG2增殖抑制作用的活性部位;抗氧化活性部位通过DPPH·清除试验筛选。结果:A、B、C、D、E 5个部位皆有一定的抗肿瘤活性和抗氧化活性,其中抗肝癌活性最强的是部位D,样品质量浓度为2.5 mg·mL^(-1)时,部位D又对人肝癌细胞HepG2增殖抑制率为74.27%;抗氧化活性最强的是部位E,当质量浓度为0.2 mg·mL^(-1)时,其对自由基DPPH清除率高达85.78%。结论:文冠果叶具有对人体有益的功效,适合开发为保健茶、化妆品等。 Objective: To study the antioxidant and antitumor activity of different extracts of Xanthoceras sorbifolia leaves. Methods: Water, 30%, 50%, 70%, and 95% ethanol aqueous were used to extract leaves of X. sorbifolia, and five different fractions A, B, C, D and E were obtained, MTT method was used to screen the antitmnor part on the proliferation of human hepatoma cell line HepG2; antioxidant parts was screed by DPPH radical-scavenging ratiorr Results: Five different fractions all showed certain antitumor and antioxidant activity, part D with the strongest anti-tumor activity, when the sample concentration was 2.5 mg·mL^-1, its inhibition rate could up to 74.27%; the strongest antioxidant activity part was E. When the concentration was 0. 2 mg·mL^-1, the free radical DPPH radical scavenging rate was as high as 85. 78%. Conclusion: The leaf of X. sorbifolia with beneficial effects for human beings, is suitable to develop products as health tea and cosmetics.
