柚皮素对高脂饮食肥胖大鼠下丘脑神经肽Y和刺鼠色蛋白相关蛋白mRNA表达的影响 被引量:1

摘要 目的观察柚皮素对高脂饮食肥胖大鼠下丘脑神经肽Y(NPY)和刺鼠色蛋白相关蛋白(AGRP)mRNA表达的影响。方法 SD大鼠44只随机分为正常对照组、高脂饮食组、高脂饮食+柚皮素低、中、高剂量组[柚皮素剂量分别为12.5、25.0及50.0 mg/(kg·d)]每天定时灌胃,记录进食量。8周末采血测血清瘦素,并取下丘脑,提取下丘脑总RNA,通过实时荧光定量聚合酶链反应(RT-PCR)测定NPY和AGRP mRNA的含量。结果给予高脂饮食后大鼠血清瘦素增高,下丘脑NPY和AGRP mRNA的表达增高,高脂饮食大鼠给予柚皮素后,血清瘦素水平降低,下丘脑NPY和AGRP mRNA的表达降低。结论柚皮素可明显降低血清瘦素水平,降低下丘脑NPY和AGRP mRNA的表达。 【Objective】 To study the effect of Naringenin on mRNA levels of neusopeptide Y(NPY) and agouti-related protein(AGRP) in hypothalamus of high fat diet obese rats. 【Methods】 Forty-four healthy male SD rats were randomly divided into five groups: control group, high fat diet group, high fat diet plus low,medium and high dose of Naringenin group(The dosages of Naringenin are 12.5, 25.0 and 50.0 mg/(kg·d)respectively). Time to fill the stomach every day. Food intake was monitored daily. Eight weeks later, the rats were sacrificed to collect blood for the measure of the levels of serum Leptin of high fat diet obese rats. Real time Polymerase Chain Reaction(PCR) was used to measure the relative mRNA levels of neusopeptide Y(NPY) and agouti-related protein(AGRP) in hypothalamus. 【Results】 Compared with control group, the rats in high fat diet group displayed increased the levels of serum Leptin, the relative mRNA levels of neusopeptide Y(NPY) and agouti-related protein(AGRP) in hypothalamus. Naringenin lowered the levels of serum Leptin,the relative mRNA levels of neusopeptide Y(NPY) and agouti-related protein(AGRP) in hypothalamus in rats fed with high fat diet. 【Conclusion】 Naringenin can reduce serum Leptin, the relative mRNA levels of neusopeptide Y(NPY)and agouti-related protein(AGRP) in hypothalamus.
