摘要
目的前期研究显示软皮汤能改善硬皮病小鼠疾病模型皮肤纤维化程度,本研究进一步探讨软皮汤对纤维化重要通路TGF-β途径的活化因子SMAD3和抑制因子FLI1的影响。方法构建硬皮病小鼠疾病模型,采用Masson三色染色皮肤组织,Western Blot法检测皮肤CollagenⅠ、Fibronectin、p-SMAD3和FLI1表达水平。结果 (1)Masson染色结果显示,模型对照组动物皮肤中胶原纤维含量明显高于正常对照组(P<0.05);软皮汤可减少软皮汤处理组小鼠皮肤中胶原纤维含量。(2)Western Blot结果显示,模型对照组小鼠皮肤中CollagenⅠ、Fibronectin、p-SMAD3的表达水平高于正常对照组,而FLI1的表达则低于正常对照组(P<P0.05);软皮汤处理组小鼠皮肤中CollagenⅠ、Fibronectin、p-SMAD3的表达水平低于模型对照组,而FLI1的表达则高于模型对照组(P<0.05)。结论软皮汤可改善硬皮病小鼠模型皮肤组织纤维化程度,其机制可能与下调活化因子p-SMAD3、上调抑制因子FLI1有关。
Objective To further study the impacts of Ruanpi decoction on activating factor SMAD3 and inhibiting factor FLI1 of TGF-β pathway as the preliminary study showed that Ruanpi decoction could relief skin fibrosis of scleroderma mouse models. Methods Build scleroderma mouse models and detect the expression level of skin Collagen I, Fibronectin and p-SMAD3 FLI1 by Masson trichrome staining skin tissue and Western Blot. Results 1.Scleroderma mouse models were built successfully. Masson staining result showed that collagenous fiber content in animal skin of model control group was obviously higher than that in normal control group(P〈0.05); Ruanpi Decoction could reduce collagenous fiber content in mouse skin in Ruanpi Decoction treatment group. 2. Western Blot result showed that the expression level of Collagen I, Fibronectin and p-SMAD3 in model control group was higher than that in the normal control group, but the expression level of FLI 1 was lower than that in the normal control group(P〈0.05); the expression level of Collagen I, Fibronectin and p-SMAD3 with Ruanpi decoction was lower than that in model control group, but the expression level of FLI1 was higher than that in model control group(P〈0.05). Conclusions Ruanpi decoction could relief skin fibrosis of scleroderma mouse models,which mechanism might be associated with the down-regulating activating factor p-SMAD3 and up-regulating inhibiting factor FLI 1.
