摘要
目的:探讨复方茵柏颗粒对四氯化碳(CCl4)致肝损伤大鼠TNF-α及细胞凋亡的影响。方法:大鼠腹腔单次注射40%CCl4玉米油制备急性肝损伤模型,将大鼠随机分为6组:正常对照组、模型组、复方茵柏颗粒低、中、高剂量组(2.16 g·kg-1、4.32 g·kg-1、8.64 g·kg-1)、阳性对照组(复方茵柏合剂8.64 g·kg-1),用酶联免疫吸附法测定血清TNF-α水平,用TUNEL法检测组织凋亡细胞,用酶联免疫吸附法检测肝匀浆Caspase-3的活性。结果:复方茵柏方能明显降低肝损伤组大鼠血清TNF-α水平,抑制细胞凋亡,并明显降低肝匀浆Caspase-3的蛋白表达量,与模型组比较有显著差异(P<0.01,P<0.05)。结论:复方茵柏颗粒抗CCl4所致肝损伤的作用与抑制TNF-α分泌、抑制细胞凋亡有关。
Objective:To investigate the effects of complex prescription Yinbai Granule on TNF- α and hepatic cell apoptosis with acute liver injury induced by carbon tetrachloride in rats. Methods:Acute liver injury was induced by 40%carbon tetrachloride in rats. Rats were randomly divided into 6 groups: normal control group,model group,complex prescription Yinbai Granule low dosage,middle dosage and high dosage groups(2. 16 g·kg^-1,4. 32 g·kg^-1,8. 64 g·kg^-1),positive control(complex prescription Yinbai mixture 8. 64 g·kg^-1). Enzyme- linked immunosorbent assay(ELISA) was used to detect the TNF-α content in serum and Caspase-3 protein expression in liver homogenate. The apoptosis index(AI) of hepatic cell was measured by TUNEL method. Results:After being treated by complex prescription Yinbai Granule,the serum level of TNF- α and apoptosis index was decreased obviously in rat with liver injury induced by CCl4(P〈0.01,P〈0.05). Caspase-3 protein expression in liver homogenate was decreased significantly(compared with liver injury group,P〈0. 01). Conclusion:Complex prescription Yinbai Granule has protection effect on acute hepatic injury in rats induced by carbon tetrachloride and the mechanism is related to inhibiting TNF- α and cell apoptosis.
