摘要
目的探讨补中益气汤对肺腺癌顺铂耐药细胞株A549/DDP裸鼠移植瘤的作用机制。方法 60只裸鼠随机分为6组,空白对照组给予生理盐水0.5ml灌胃,其余5组以5×106/L的A549/DDP接种于裸鼠皮下,待成瘤后,分别给予生理盐水0.5ml灌胃,0.2ml顺铂腹腔注射同时生理盐水灌胃,0.2ml顺铂腹腔注射顺铂和高、中、低剂量的补中益气汤灌胃。观察A549/DDP细胞在各组裸鼠的致瘤率,计算抑瘤率;采用免疫组化方法、RT-PCR(real time PCR)检测移植瘤组织中PI3K、AKT的表达水平。结果补中益气汤高、中、低剂量组能不同程度的抑制移植瘤的生长,其中以高、中剂量组最显著,与顺铂组抑瘤率比较有明显的统计学差异(P<0.05);移植瘤中PI3K、AKT蛋白表达和mRNA基因的表达在补中益气汤高、中、低剂量组中均有下降,高、中剂量组降低较多,与顺铂组和小剂量比较有统计学差异(P<0.05)。结论补中益气汤联合顺铂对肺腺癌移植瘤的增殖有明显的抑制作用,补中益气汤能通过调控PI3K、AKT基因和蛋白的表达而干预顺铂的耐药机制,这种干预作用具有一定的量效关系。
Objective To explore the effects and mechanisms of Buzhong Yiqi Decoction on transplantation tumor of human lung adenocarcinoma metastases with cisplatin-resistant cell（ A549/DDP） in nude mice. Methods 60 nude mice were randomly divided into 6 groups and given corresponding intervention measurees. After 14 days,to observe the tumorigenic rate and calculate tumor inhibition rate,use immunohistochemical method and rt-PCR（ real time PCR） method to detect the expression level of PI3 K,AKT in transplantation tumor tissue. Results The different dose of Buzhong Yiqi Decoction can inhibit the growth of transplantation tumor. High and mederate dose groups are the most significant. Comparing with cisplatin group,there are statistically significant（ P 0. 05）; PI3 K and AKT protein expression and gene expression in high,mederate and low dose groups were decreased. There are statistically difference between high,mederate dose groups and cisplatin and small dose groups（ P 0. 05）.Conclusion The Buzhong Yiqi Decoction combined with cisplatin has obvious inhibitory effect in lung adenocarcinoma metastases; The Buzhong Yiqi Decoction can intervene cisplatin resistance mechanism throught regulating the gene and protein expression of PI3 K,AKT. The intervene has certain dose-effect relationship.
