摘要
目的观察肉苁蓉苯乙醇苷预防给药对高原脑水肿模型大鼠的作用及可能的作用机制。方法大鼠随机分为正常对照组,模型组,大花红景天口服液组[1.78 mL/(kg·d)],肉苁蓉苯乙醇苷低、中、高剂量组[75、150、300 mg/(kg·d)],每组12只,灌胃预防给药10 d,第8日起除正常对照组外其余各组置于模拟海拔5 000 m高原环境72 h建立高原脑水肿模型,观察各组大鼠脑组织病理改变,检测脑组织含水量、脑组织匀浆中白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)酶活力。结果与正常对照组相比,模型组大鼠出现明显的脑水肿,脑组织含水量显著升高,脑匀浆液中IL-6、TNF-α、MDA含量增高,SOD和GSH-Px酶活力显著降低。肉苁蓉苯乙醇苷能够改善高原脑水肿大鼠脑水肿病理改变,降低脑组织含水量,降低脑匀浆中IL-6、TNF-α、MDA含量,且能提高SOD和GSH-Px的酶活力。结论肉苁蓉苯乙醇苷能够预防高原脑水肿的发生,可能与其抗炎、抗氧化应激有关。
Objective To observe the effect and mechanism of phenylethanoid glycosides from Cistanche Salsa on rats with high-altitude cerebral edema. Methods Phenylethanoid glycosides were given prophylactically to establish the rat model of high-altitude cerebral edema by using hypobaric chamber in a simulated environment of 5 000 m altitude. Pathologic changes in the rat lung tissue, content of water, IL-6, TNF-α, MDA and enzymatic activity of SOD and GSH-Px in the lung homogenate were measured. Results Compared with the control group, the high-altitude cerebral edema rats showed remarkable character of cerebral edema and high content of water in the lung. The content of IL-6, TNF-α and MDA in the lung homogenate was increased while enzymatic activity of SOD and GSH-Px was signifi cantly decreased. The phenylethanoid glycosides could improve the pathological changes in the lung tissue, decrease the content of water, IL-6, TNF-α and MDA, while increase the enzymatic activity of SOD and GSH-Px in the lung homogenate. Conclsuion Phenylethanoid glycosides can prevent high-altitude cerebral edema, whose mechanism may related to the anti-infl ammation and anti-oxidatiin stress in the lung tissue.
