摘要
肉苁蓉细胞悬浮培养的生长周期约为25 d,细胞内苯乙醇糖甙(PeG)的比生成速率YP/X与肉苁蓉细胞比生长速率μ大致成线性关系,为生长偶联型.在培养过程中,培养基中的磷酸盐和氨盐被迅速消耗,而硝酸盐的利用稍慢,基于蔗糖、氨盐和硝酸盐的细胞收率系数分别为0.481、60和6.4 g/g,基于蔗糖的PeG收率系数为0.055 g/g.根据细胞基本元素分析及各收率系数之间的关系,计算得出了肉苁蓉细胞悬浮培养的化学计量方程式,为细胞的生物反应器扩大培养提供理论依据.
The growth period of Cistanche deserticola cells in suspension culture is about 25 days.The relationship between specific productivity of phenylethanoid glycosides and cell specific growth rate growth behaved linearly.During the growing period,phosphate and ammonia in culture media were consumed quickly, but nitrate was used slowly.The cell yield coefficients based on sucrose,ammonia and nitrate were 0.48,160 and 6.4 g/g respectively.PeG yield coefficient based on sucrose was 0.055 g/g.The stoichiometrical equation of C.deserticola cells in suspension culture was deduced by the analysis of elemental composition of cells and these yield coefficients.
