摘要
以苹果清汁为样品,优化多酚检测条件,建立了不同品种苹果清汁中多酚的HPLC指纹图谱.结果表明,采用Welchrom C18色谱柱,在检测波长280 nm,柱温30℃,进样量20μ1,流动相0.3%三氟乙酸水溶液和甲醇(流速1ml/min)以及梯度洗脱的条件下,标定了12个共有指纹峰,测定出10个品种苹果清汁多酚的指纹相似度均在0.900以上,多酚含量为1 974 ~3 598 μg/ml,初步建立了苹果清汁的高效液相指纹图谱.该方法可为苹果加工企业对苹果多酚的质量进行定性、定量地控制提供试验依据.
The HPLC fingerprint of polyphenols in different varieties apple juice was established using apple juice as a sample and after optimizing the polyphenol analysis conditions.The results show that under those conditions of using Welchrom C18 column,at the detection wavelength of 280 nm,the column temperature 30 ℃,amount of sample 20 μl,mobile phase of 0.3% three trifluoroacetic acid aqueous solution and methanol (flow 1 ml/min),gradient elution,12 common fingerprint peaks were determined.As well as the finger print similarity of polyphenol is above 0.900,and the content of polyphenol in 10 categories apple juice was 1 974 to 3 598 μg/ml.So the HPLC fingerprint of apple juice was initially established.This method will provide the test basis for apple juice processing enterprises in their qualitative and quantitative analysis and control of apple polyphenol.
