摘要
目的:研究子宫内膜异位症(EMs)患者在位、异位内膜干细胞分离、培养及鉴定方法。方法:采用胶原酶III法进行体外分离、培养子宫内膜异位症患者在位、异位内膜干细胞,倒置显微镜下观察贴壁细胞的生长情况,并运用免疫荧光和RT-PCR技术检测干细胞相关标记物的表达。结果:成功培养出悬浮生长的子宫内膜干细胞球,CD34、CD117抗原及Oct-4因子在子宫内膜细胞中呈持续的阳性表达。结论:EMs患者在位、异位内膜中存在干细胞,对其分离、培养,可作为研究EMs发病机制的体外模型。
Objective : To study stem cell isolation, culture and identification of endometriosis (EMs) patients. Methods : By using collagenase III, stem cells of endometriosis were isolated and cultured in vitro. The growth of cells was observed under inverted microscope and by using immunofluorescence and RT - PCR technique, the expression of stem cells related markers was detected. Results:The suspended growth of endometrial stem cells was successfully developed. CD34, CD117 antigen and Oct -4 factor in endometrial cells continued to have positive expression. Conclusion:In patients with EMs, stem cells exist in the ectopic endometrium. The isolation as well as culture can be used for research in vitro.
