摘要
目的采用HPLC测定麻黄汤组方变化时对麻黄碱和伪麻黄碱煎出量的影响。方法制备麻黄汤,用均匀实验设计的16个不同配比麻黄汤水煎液,采用HPLC法测定麻黄碱和伪麻黄碱的含量,计算其与麻黄单独煎煮相比的煎出率;采用逐步回归法分析处方组成对麻黄碱和伪麻黄碱煎出量的影响。结果所用HPLC测试方法分离效果好,灵敏度高、重复性好,不同配比麻黄汤水煎液中麻黄碱和伪麻黄碱的含量有明显差异。结论麻黄碱和伪麻黄碱煎出量具有极显著的正相关性;杏仁能极显著地增加麻黄碱、伪麻黄碱在水煎液中的煎出量。
OBJECTIVE To establish a quantitative method for ephedrine and pseudoephedrine in the aqueous extract of Mahuang deeoction(MHD) by HPLC,and to evaluate the influence of the aqueous extract from MHD with different ratios on the decocting contents of ephedrine and pseudoephedrine by HPLC method. METHODS Aqueous extracts of MHD with 16 different ratios was prepared via uniform experimental design,and then the contents of ephedrine and pseudoephedrine was determined by HPLC method, and thereby calculated the decocting rates of ephedrine and pseudoephedrine of these above 16 formulas and decoction from Ephedrae herba alone. Subsequently, the influence of different compositions of prescriptions was analyzed on the decocting contents of ephedrine and pseudoephedrine by stepwise regression method. RESULTS I-IPLC method in this study was good in the aspect of separable effect, sensitivity and reproducibility. And the contents of ephedrine and pseudoephedrine of the aqueous extract from Mahuang decoction with different ratios were significant different. CONCLUSION There is a highly significant positive correlation between the decocting contents of ephedrine and pseudoephedrine. And Armeniacae amarum semen can obviously increase the decocting contents of ephedrine and pseudoephedrine of MHD.
