肠球菌属益生菌提取物LFK对变应性鼻炎小鼠血清IL-12的影响 被引量:6

Effect of lysed Enterococcus faecalis FK-23(LFK) on serum IL-12 in allergic rhinitis mice

摘要 目的变应性鼻炎(allergic rhinitis,AR)小鼠模型经口途径给予经热处理及酶裂解的肠球菌faecalis FK-23菌株(lysed Enterococcus faecalis FK-23,LFK),观察对血清IL-12水平的影响,探讨LFK对实验性AR的抗变态反应作用。方法 BALB/c小鼠75只,随机分为阳性对照组、LFK干预组和阴性对照组。采用OVA腹腔注射及鼻腔激发建立小鼠AR模型,阴性对照组小鼠采用生理盐水代替OVA进行腹腔注射及滴鼻。用益生菌提取物LFK0.5 mL(60 mg)对AR小鼠模型进行灌胃,共计21 d;阳性和阴性对照组以生理盐水0.5 mL代替LFK灌胃处理。第22天取材,用酶联免疫吸附试验(ELISA)检测小鼠血清IL-12水平。结果血清IL-12水平阳性对照组较阴性对照组显著降低(P<0.001),LFK干预组较阳性对照组显著升高(P<0.001),但与阴性对照组相比仍明显降低(P<0.001)。结论肠球菌属益生菌提取物LFK对细胞因子IL-12具有一定的调节作用,推测其与抗变态反应作用有关。 Objective To observe the effect of the heat-treated and lysed Enterococcus faecalis FK-23 ( LFK) on serum levels of IL-12 in a murine model of allergic rhinitis ( AR) , and to discuss the anti-allergic effect of LFK on experimen-tal AR.Methods A total of 75 BALB/c mice were divided into three groups:1 ) positive control group, 2 ) LFK-treated group, and 3) negative control group.To establish the murine AR model, intraperitoneal injection and nasal drip with ovalbumin ( OVA) were performed.Saline was used instead of OVA for negative control mice.Probiotic prepara-tion of LFK was orally administrated in a process of 21 days (60 mg/0.5 mL/mouse per day) in LFK-treated mice. The positive and negative control mice received saline orally for 21 days.The IL-12 levels were quantified in serum using ELISA on day 22.Results Serum levels of IL-12 were significantly decreased in the positive control group than those in the negative control group (P〈0.001), and significantly increased in the LFK-treated group than those in the positive control group (P〈0.001).However, serum levels of IL-12 were still significantly decreased in the LFK-trea-ted group compared to the negative control group (P〈0.001).Conclusion LFK, an Enterococcus probiotic prepara-tion, has certain modulatory actions on cytokine IL-12, which might be related to its anti-allergic effect.
