摘要
目的 为探讨细脚拟青霉总多糖对大鼠机体免疫功能的调节作用 .方法 用细脚拟青霉总多糖 (10 0 mg/ kg体重 )臀部皮下注射处理大鼠 8天后 ,测定大鼠外周血白细胞数和血红蛋白含量 ;用含 10 %小牛血清的 RPMI- 16 40细胞培养液贴壁培养肺泡和腹腔巨噬细胞 2 h后 ,用全自动生化分析仪测定其中乳酸脱氢酶与酸性磷酸酶活性 ,并进行中性红吞噬试验 .结果 细脚拟青霉总多糖使大鼠肺泡和腹腔巨噬细胞内乳酸脱氢酶和酸性磷酸酶的活性均升高 (P<0 .0 0 1;P<0 .0 0 1;P<0 .0 1;P<0 .0 1) ,对中性红摄取能力也均增强 (P<0 .0 0 1;P<0 .0 0 1) .结论 细脚拟青霉总多糖对大鼠肺泡和腹腔巨噬细胞具有激活作用 。
Objective To study the immune regulation of paecilomyces tenuipes total polysaccharides on the rats.Methods After subcutaneous injection with paecilomyces tenuipes total polysaccharides(100mg/kg weight) in the back of the rats for 8 days, the white blood cell counts (WBC) and the levels of haemoglobin (Hb) in the blood of rats were determined; and after the alveolar macrophages (AMΦ) and peritoneal macrophages (PMΦ) cultured with 10% Fetal Bovine Serum RPMI 1640 for 2h in vitro, the activities of lactate dehydrogenase (LDH) and acid phosphatase (ACP) in the AMΦ and PMΦ were detected by automatic biochemistry analyzer, and the abilities devouring neutral red in the AMΦ and PMΦ were observed.Results The activities of ACP and LDH were increased and the abilities devouring neutral red were strengthened in the AMΦ and PMΦ of rats induced by paecilomyces tenuipes total polysaccharides. Conclusion Paecilomyces tenuipes total polysaccharides can activate the AMΦ and PMΦ and heightene non specificity immunity of rats.
