摘要
目的研究肝刺激物(hepatic stimulator substance,HSS)对人肝癌细胞凋亡的影响。方法 用无血清培养人肝癌细胞诱导典型的细胞凋亡,以此为模型应用流式细胞术及流式细胞免疫荧光技术,检测HSS作用不同时间后,对无血清培养人肝癌细胞诱导凋亡比率的影响,并观察HSS对常规培养人肝癌细胞的P53蛋白表达变化的影响。结果HSS作用于无血清培养的人肝癌细胞,细胞凋亡比率随时间延长而增加;HSS作用于常规培养的人肝癌细胞,细胞 P53蛋白的表达随时间延长而减弱。结论 HSS对无血清培养人肝癌细胞凋亡有促进作用。
To study the effect of hepatic stimulator substance (HSS) on the apop-tosis of human hepatoma cells. Methods Apoptosis was induced by serum-deprivation and was demonstrated in morphological observation. Flow cytometry and cellular immunofluorescence technique assayed the rates of apoptosis and the expressions of P53 protein of human hepatoma cells of SMMC-7721 cell lines treated by HSS(50μg/ml) in different times. Results The rate of apoptosis of hepatoma cells induced by serm-deprevation was increased and the expression of P53 of hepatoma cells was decreased when HSS was administrated. Conclusion HSS promotes the apoptosis of hepatoma cells.
