摘要
目的:研究肝细胞刺激因子(HSS)与阿霉素(ADM)联合用药对裸鼠肝癌移植瘤细胞的凋亡作用。方法:建立裸鼠肝癌移植瘤模型;将荷瘤裸鼠分组,分别腹腔内注射不同剂量HSS及ADM;4周后解剖取瘤计算抑瘤率;病理组织学检查;采用AnnexinV/PI流式细胞分析法检测肝癌细胞凋亡率。结果:与对照组相比,三种剂量联合用药组抑瘤作用明显(P<0.01)。HSS组诱导细胞凋亡作用不明显(P>0.05),ADM组细胞凋亡作用明显(P<0.01),但裸鼠有明显的不良反应。中小剂量联合用药组细胞凋亡率明显增加(P<0.01),与ADM组相比也表现出明显差异(P<0.01)。大剂量联合用药组诱导细胞凋亡作用明显(P<0.01),与ADM组相比差异同样有显著性(P<0.01),但裸鼠表现出强烈的毒副反应。结论:中小剂量HSS和ADM联合用药抑瘤作用显著,对诱导裸鼠肝癌细胞的凋亡具有协同作用。
Objective To study the co-effect of hepatic stimulator substance(HSS) in combination with adriamycin(ADM) on inducing the cellular apoptosis in xenograft hepatocellular carcinoma in nude mice.Methods SMMC-7721 was implanted subcutaneously to the posterior regions of 48 nude mice to establish the transplantation tumor model of human liver cancer.ADR + HSS was injected into the peritoneal cavity of nude mice with xenograft hepatocellular carcinoma.The pathological features of tumor tissues and some other important organs were examine,and the tumor growth inhibitory rate was calculated.FITC-Annexin V /PI were used for measuring apoptosis of the tumor cells by flow cytometry.Results HSS in combination with ADM inhibited the tumor growth significantly(P〈0.01).HSS alone had no effect in inducing the cellular apoptosis(P〉 0.05).ADM induced the cellular apoptosis significantly(P 〈0.01),but resulted in severe side effect.The rate of apoptosis induced by small and middle dosage of ADM(1 mg /kg or 2 mg /kg) + HSS(1 mg /kg)was increased significantly(P〈 0.01),which was significantly different from that of ADM(P〈 0.01).Large dosage of ADM(4 mg /kg) + HSS(1 mg /kg) induced the cellular apoptosis significantly(P 〈0.01),but still resulted in side effect severely.Conclusion HSS in combination with ADM might improve the tumor inhibitory effect significantly,and the co-effect could promote the cellular apoptosis in nude mice with xenograft hepatocellular carcinoma.
