摘要
目的 了解长期饮酒对人体组织细胞损伤的可能原因。方法 采用不同浓度酒精加入体外培养的人红白血病(K562)细胞中,用荧光显微镜和倒置光显微镜观察细胞形态学变化,用琼脂糖凝胶电泳的方法检测细胞DNA裂解。结果 1%酒精能有效诱导K562细胞凋亡,且随剂量增大凋亡时间缩短。5%酒精作用30h则可引起细胞坏死。结论 长期饮酒者血液中有核细胞数量减少,可能与其促凋亡作用有关。
[Objective] To study the damage of alcohol on human k562 cells. [Methods] Alcohol of different density was added into the medium in which K562 cells were cultured. The changes of cell morphology were observed by phase contrast microscope and fluorescent light microscope. The apoptosis was detected by agarose gel electrophoresis. [Results] 1% of alcohol could efficiently induce apoptosis of K562 and latency of apoptosis was shortened with the increase of alcohol dose. 5% alcohol may cause necrosis. [Conclusion] There is a reduction of the number of nuclear cell in the blood due to long - time alcohol intake, which may be relevant with its death - promoting function.
