摘要
为了解乙醇导致血液粒细胞、单核细胞、淋巴细胞数量减少的原因,以HL-60细胞为凋亡研究模型,采用流式细胞术和电子显微术等方法探讨了乙醇对细胞凋亡的影响。结果表明,HL-60细胞在50、100、150、200、250m m ol/L乙醇处理72h 后,各组凋亡细胞比例均比对照组显著增高(P< 0.01),且呈剂量-效应关系(r= 0.975)。说明乙醇对HL-60细胞有促凋亡作用。提示长期饮酒者粒细胞、单核巨噬细胞。
The purpose of this study was to examine the effect of ethanol on apoptosis of human promyelocytic leukemia cell line HL 60 cells and search for a possible reason for reduced PMNs、monocytes、lymphocytes in the blood of alcoholics. Human promyelocytic leukemia cell line HL 60 cells was used as a model of apoptosis, with ethanol being the interfering factor. Qualitative and quantitative detection was done by both electron microscopy and flow cytometry. Compared with the control treatment of HL 60 cells with ethanol at 50,100,150,200,250mmol/L for 3 days increased apoptotic cells significantly(P<0 01), and apoptosis was dose dependent for ethanol. It suggest that ethanol can induce apoptosis of HL 60 cells. So as far as reduced PMNs, monocytes, phagocytes and lymphocytes of alchoholics are concerned, ethanol induced apoptosis may well be an important reason.
