摘要
目的 研究脑出血大鼠缺血半暗带内HSP70与NF κB的表达及FNS的作用。方法 采用Ⅶ型胶原酶法大鼠脑出血模型。分别预刺激小脑齿状核与小脑顶核 ,观察血肿周围水肿区尼氏染色、HSP70与NF κBP6 5免疫反应阳性细胞光密度。结果 预刺激小脑顶核可使行为计分降低 ,死亡神经细胞数目与毛细血管周围间隙面积减少 ,尼氏染色与HSP70反应阳性细胞光密度增高 ,NF κBP6 5光密度减少 ,预刺激小脑齿状核无此作用。结论 预刺激小脑顶核能促进大鼠神经功能恢复 ,减轻脑水肿与神经细胞破坏 ,减少神经细胞死亡 ,促进HSP70表达 ,抑制NF κB的表达。预刺激小脑顶核有抑制炎症因子的作用 ,可能是其脑保护作用机制之一。
Objective To investigate the expression of HSP70 and NF κB in ischemic Penumbra effects after experimental cerebral haemorrhage and the effects of fastigial nucleus stimulation.Methods Haemorrhagic stroke rat model was made by Collagenase Ⅶ injection method,cerebellar dentate nucleus and fastigial nucleus were stimulated separately, Nissl's staining, immunohistochemistry were taken to estimate optical densit of positive cell of Nissl's staining, HSP70 and NF κBP65 immunohistochemistry reaction in edema area around hematom.Results Fastigial nucleus stimulation could reduce the behavior scores, decrease the number of death neural cells and peri interspace area of blood caillary, increase the optical densit of positive cell of Nissl's staining, HSP70 immunohistochemistry reaction, reduce the optical densit of NF κBP65. Dentate nucleus stimulationl showed no such results.Conclusion Fastigial nucleus stimulation could promote the functional restoration of rats, relieve brain edema and nerve cell destruction, decrease nerve cell death, promote the expression of HSP70 and inhibit the expression of NF κB. Fastigial nucleus stimulation had the function to inhibit the inflammation reaction and that could be one of its mechanisms to protect brain.
