红芪多糖HG-2含量及联合透明质酸水凝胶的参数测定 被引量:4

摘要 目的制备红芪多糖HG-2及建立红芪多糖HG-2的含量测定方法,并联合透明质酸水凝胶测定凝胶时间、释药量、降解及溶胀率等参数。方法采用SephadexG-25纯化制备红芪多糖HG-2,苯酚-硫酸法测定含量,并用化学改性法制备透明质酸(HA)水凝胶,将二者联合制备红芪多糖HG-2HA水凝胶。结果红芪多糖HG-2的含量为95.97%,HA水凝胶的凝胶时间为(180±20)s;溶胀率为42.33%;并在第5天基本降解。红芪多糖HG-2HA水凝胶释药在第120h已趋于平稳,释药量为86.15%。结论选用葡萄糖作为对照品测定红芪多糖HG-2含量的方法简便、准确。红芪多糖HG-2HA水凝胶能够起到缓释作用,二者联用具有可行性,可以为后续动物实验提供理论依据。 Objective To prepare hedysarum polysaccharide HG-2,establish a content determination method for hedysarum polysaccharide HG-2,and to determine the gel time,release amount,degradation and swelling rate and other parameters in combination with hyaluronic acid hydrogel(HA hydrogel).Methods Hedysarum polysaccharide HG-2 was purified by Sephadex G-25.The content was determined by phenol-sulfuric acid method.HA hydrogel was prepared using the chemical modification method.The two hydrogels were combined to prepare hedysarum polysaccharide HG-2 HA hydrogel.Results The content of HG-2 in Hedysarum polysaccharide was 95.97%,the gel time of HA hydrogel was(180±20)s,the swelling rate was 42.33%,and it was basically degraded in 5 d.Release of astragalus polysaccharide HG-2 HA hydrogel was stable at 120 h,and the release rate was 86.15%.Conclusion Glucose can be used as a reference substance to determine the content of HG-2 in Hedysarum polysaccharides.Astragalus polysaccharide HG-2 HA hydrogel can play a sustained-release role.The combination of the two is feasible and can help guide subsequent experiments.
