摘要
目的比较Garcinol及其新衍生物8-Allyl Garcinol对人口腔鳞癌细胞系SCC15生物学行为的影响。方法培养人口腔鳞癌细胞系SCC15,采用不同浓度的Garcinol、8-Allyl Garcinol处理细胞后,通过MTT法检测对细胞增殖的影响、克隆形成实验检测对细胞克隆形成能力的影响、划痕实验检测对细胞迁移能力的影响、Annexin V-FITC/PI双染色流式细胞术检测对细胞凋亡的影响,比较Garcinol和8-Allyl Garcinol对人口腔鳞癌细胞生物学行为影响的差异。结果 Garcinol和8-Allyl Garcinol均能抑制人口腔鳞癌细胞SCC15的增殖、克隆形成及迁移能力,并促进细胞的凋亡,呈一定的时间和剂量依赖性。其中,8-Allyl Garcinol对细胞增殖及细胞克隆形成的抑制作用优于Garcinol(P <0. 01),但促进细胞凋亡效果较Garcinol弱(P <0. 05),对细胞迁移能力的抑制作用二者未见统计学差异(P> 0. 05)。结论 8-Allyl Garcinol对人口腔鳞癌细胞SCC15具有显著的抑制作用,但作用效果和机制与Garcinol有一定差异。
Objective To evaluate the effects of 8-Allyl Garcinol,a derivative of Garcinol,on the biological behavior of human oral squamous cell carcinoma (OSCC)cell line SCC15.Methods OSCC cell line SCC15 was cultured and treated with different concentrations of Garcinol or 8-Ally Garcinol respectively.The effects on the biological behavior of OSCC cell line SCC15 were measured.The cells proliferation was measured by MTT assay.Clone formation was examined by clonoe formation assay.Cells migration ability was measured by scratch migration assay.Cells apoptosis was observed by flow cytometer with Annexin V-FITC/PI staining assay.Results Both Garcinol and 8-Allyl Gareinol showed inhibitory effects on the proliferation,clone formation,migration ability of SCC15 and promoted cell apoptosis with certain time and dosage dependency.8-Allyl Garcinol had greater inhibitory effect on cell proliferation and clone formation than Garcinol (P<0.01),but had relatively weaker effect on promotion of apoptosis (P<0.05).There was no significant difference in the inhibitory effect on cell mobility(P>0.05).Conclusion 8-Allyl Garcinol has prominent inhibitory effect on OSCC cell line SCC15.However,the mechanism is different from Garcinol.
