摘要
目的探讨姜黄素类似物H8改善糖尿病小鼠模型肾纤维化的作用和机制。方法选取9周龄糖尿病db/db小鼠和db/m小鼠,分为正常对照组(db/m,Con)、糖尿病组(db/db)、H8治疗组(db/db+H8),每组8只。各组自给药开始计时,第8周末处死,采集血液和肾脏组织,检测H8对血清尿素(UR)、Scr、SUA特征指标的影响;HE染色、Masson染色观察肾脏病理形态变化;RT-PCR检测小鼠肾组织中胶原蛋白Ⅰ(ColⅠ)、波形蛋白(Vimentin)mRNA的表达;Western blot检测ColⅠ、Vimentin、p-ERK、ERK的蛋白表达。结果 db/db+H8组血清UR、Scr、SUA降低(P<0.05);HE、Masson染色显示,H8能改善db/db小鼠肾脏组织炎症侵袭、胶原蛋白累积与肾纤维化;与db/db组比较,db/db+H8组的RT-PCR显示ColⅠ、Vimentin mRNA水平下降(P<0.05),Western bolt结果显示,ColⅠ、Vimentin、p-ERK蛋白表达水平也降低(P<0.05或P<0.01)。结论 H8能改善db/db小鼠肾脏纤维化,可能与抑制ERK信号通路有关。
Objective To investigate the effect and mechanism of curcumin analogue (H8)on renal fibrosis in diabetic mice. Methods Diabetic db/db mice were purchased.9weeks old mice were divided into 3 groups : normal control group (Con,n =8), diabetic group (db/db,n =8)and H8treatment group (db/db+H8,n =8).All the mice were sacrificed at the end of the eighth week.The blood samples and kidney tissues were collected.The effects of H8on serum urea (UR), creatinine (Scr)and uric acid (SUA)were examined.Renal pathological changes were observed by HE staining and Masson staining. RT-PCR was used to detect the expressions of collagen typeⅠ (ColⅠ)and Vimentin mRNA.Western blot was used to detect the expressions of ColI,Vimentin,p-ERK and ERK protein in kidney tissue. Results The levels of serum UR,Scr and SUA in diabetic mice treated with H8were decreased (P<0.05).HE staining and Masson staining showed that H8could ameliorate inflammation invasion,collagen accumulation and renal fibrosis in db/db mice.Compared with db/db group,the levels of ColI and Vimentin mRNA decreased in db/db+H8group (P<0.05), the expressions of ColⅠ,Vimentin and p-ERK protein were also decreased (P<0.05 or P<0.01). Conclusion H8can attenuate renal fibrosis in db/db mice,which may be related to the inhibition of ERK signaling pathway.
