铜绿假单胞菌群体感应QS系统对Ⅰ类整合子intI1基因调控作用的初步研究 被引量:2

A preliminary exploration on regulation of the class Ⅰ integrase intI1 gene by the quorum-sensing system in Pseudomonas aeruginosa

摘要 目的研究铜绿假单胞菌Ⅰ类整合子阳性株群体感应系统基因lasR与Ⅰ类整合子整合酶基因intI1表达水平的相关性,初步探讨群体感应系统对Ⅰ类整合子的调控机制。方法通过荧光定量RT-PCR方法检测铜绿假单胞菌群体QS信号分子受体基因lasR和Ⅰ类整合子整合酶基因intI1在生物被膜中与阿奇霉素菌液中mRNA的表达水平,分析两基因表达的相关性。结果铜绿假单胞菌群体感应系统lasR基因及Ⅰ类整合子intI1基因表达水平在生物被膜中均较在阿奇霉素菌液中升高(P<0.05),且两者表达水平呈正相关性(r=0.565,P<0.05)。结论铜绿假单胞菌群lasR和intI1基因表达呈正相关,且阿奇霉素对Ⅰ类整合子intI1基因表达抑制作用明显,提示intI1基因表达可能与群体感应系统的调控作用有关。 OBJECTIVE The regulation mechanism of the quorum-sensing system on the class 1 integron were preliminarily explored by studying the expression correlation between the quorum-sensing system lasR gene and the class 1 integron integrase intⅠ1 gene of Pseudomonas aeruginosa.METHODS Fluorescence quantitative RT-PCR was used for detecting the mRNA expression level of the quorum-sensing system signal molecular receptor lasR gene and the class 1 integron integrase intⅠ1 gene of P.aeruginosa in biofilm and azithromycin-supplemented bacterial solution,and the correlation between the expression of the two genes was analyzed.RESULTS In biofilm,the mRNA expression levels of P.aeruginosa quorum-sensing system lasR and the class 1integron intⅠ1 gene were both significantly greater than those in the azithromycin-supplemented bacterial solution (P<0.05),and the expression of these two genes had positive correlation (r=0.565,P<0.05).CONCLUSION The expression of lasR and intⅠ1 genes of P.aeruginosa is positively correlated and azithromycin has obvious inhibitory effect on the expression of the class 1 integrase intⅠ1 gene,which suggests that the intⅠ1 gene expression may be related to the regulation function of the quorum-sensing system.
