芪术蚤复方对乳腺癌模型小鼠的抑制作用及机制研究 被引量:3

摘要 目的:观察芪术蚤(QZZ)复方对乳腺癌模型小鼠的影响,并探讨其作用机制。方法:4T1乳腺癌细胞于右胸皮下注射,建立乳腺癌模型小鼠,取50只造模成功的模型小鼠随机分5组,每组10只,分别为模型对照组、阳性对照组、芪术蚤复方20mg/kg、40mg/kg、80mg/kg组,阳性对照组腹腔注射顺铂(1mg/kg)、芪术蚤复方20mg/kg、40mg/kg、80mg/kg组分别灌胃给予芪术蚤复方20mg/kg、40mg/kg、80mg/kg,模型对照组给予等体积生理盐水,1次/日,连续给药2周。停药第二天分离瘤体、胸腺和脾脏,计算重量、抑瘤率、胸腺指数及脾指数、观察瘤体病理学结果; ELISA检测小鼠血清IL-2、IL-6、IL-12、TNF-α水平,RT-PCR检测模型小鼠血清TGF-β1、TFN-γ、VEGF mRNA的表达水平,流式细胞仪检测各组腺癌模型小鼠外周血中CD4+、CD8+水平及值。结果:给药2周后与模型组相比,芪术蚤复方可显著抑制乳腺癌模型小鼠肉瘤的生长,提高胸腺指数和脾指数(P<0. 05);通过病理学观察发现芪术蚤复方给药组癌细胞排列疏松,核固缩,血管较模型对照组减少明显;芪术蚤复方给药组小鼠血清中IL-2、IL-6、IL-12和TNF-α的水平也显著升高(P<0. 05),TGF-β1、VEGF mRNA的表达水平显著降低(P<0. 05),小鼠外周血中CD4+/CD8+的水平显著升高(P<0. 05)。结论:芪术蚤复方可抑制肿瘤细胞生长,其机制可能是通过调节乳腺肿瘤微环境中调节性T细胞的数量及比例提高乳腺癌模型鼠免疫功能,进而抑制乳腺癌模型小鼠乳腺肿瘤组织的生长。 Objective To investigate the influence for breast cancer model and antitumor mechanism of compound Qizhuzao (QZZ),Method 4 T1 cell in right chest subcutaneously,establish a tumor-bearing mice model of breast cancer,50 successful models were randomly divided into five groups with 10 in each group:model control group,Positive control group,three different concentration (20,40,80mg ·kg -1)of QZZ groups,positive control group Intraperitoneal injection with cisplatin (1mg/ kg),three QZZ groups were infused to stomach with 20mg/kg.40mg/kg,80mg/kg,The model control group was given the equal volume of NS,once a day for consecutive 2 weeks,tumor tissue,thymus and spleen were separated after the administration, calculate the weight,inhibitory rate,the thymus index and spleen index,observed the pathological consequences,ELISA was employed to detect of cytokines interleukin-2(IL-2),interleukin-12(IL-6),interleukin-12(IL-12)and TNF-alpha, the expression of TGF-β1,VEGF mRNA in serum were detectde with rt-pcr,T cell subsets of CD4^+ and CD8^+ in peripheral blood of tumor-bearing mice were measured by flow cytometry.Result Compared with model group,QZZ groups were significantly inhibit the growth of mice sarcoma and improve the thymus index,spleen index (P<0.05),the pathological consequences shows cancer cells loosely arranged,nucleus pycnosis,blood vessels in the QZZ groups,the IL -2,IL -6,IL -12 and TNF -α were significantly increased (P<0.05)and TGF-β1,VEGF mRNA expression level were significantly decreased (P< 0.05) in the QZZ groups,also the serum CD4^+/CD8^+ were significantly increased (P<0.05).Conclusion Compound of QZZ can directly inhibit tumor cells,improve the immune function of breast cancer model mice by adjusted the number and proportion of regulatory T cells in breast tumor microenvironment,then restrain the growth of the tumor-burdened breast tumor in mice.
