前列通瘀汤对前列腺增生模型大鼠组织蛋白激酶调控及抑制前列腺增生机制影响 被引量:7

摘要 目的:分析前列通瘀汤对良性前列腺增生(BPH)大鼠组织蛋白激酶调控及抑制前列腺增生机制影响。方法:选取由本院实验动物中心提供的SPF级Wistar雄性大鼠45只,随机数字表法将大鼠分成三组,观察组(15只)、模型组(15只)和正常组(15只),模型组与观察组大鼠制备BPH模型,成功造模后观察组大鼠每天灌胃1ml/100g前列通瘀汤,模型组和正常组灌胃等剂量生理盐水,连续灌胃30天;末次给药后检测各组大鼠体重、前列腺湿重及前列腺指数状况,原位杂交法检测大鼠其前列腺组织内PKCmRNA及PKAmRNA阳性表达状况,ELISA法检测各组大鼠前列腺组织内血管内皮生长因子(VEGF)、超氧化物歧化酶(SOD)、表皮生长因子(EGF)、谷胱甘肽过氧化酶(GPx)及丙二醛(MDA)含量。结果:模型组大鼠前列腺重量及前列腺指数较正常组显著升高,观察组大鼠前列腺重量及前列腺指数较模型组显著降低,差异均有统计学意义(P<0. 05);模型组PKCmRNA阳性表达率及PKC/PKA较正常组显著升高,PKAmRNA阳性表达率较正常组显著降低,观察组PKCmRNA阳性表达率及PKC/PKA较模型组显著降低,PKAmRNA阳性表达率较模型组显著升高,差异均有统计学意义(P <0. 05);观察组和模型组EGF、VEGF、MDA含量较正常组显著升高,SOD、GPx含量较正常组显著降低,观察组升高、降低幅度低于模型组,差异均有统计学意义(P<0. 05)。结论:前列通瘀汤可改善BPH大鼠前列腺组织内蛋白激酶失衡状况,对前列腺增生起到抑制作用,同时可调节大鼠机体内血管生长和氧化应激程度。 Objective:To analyze the effect of Qianlie Tongyu Decoction on the regulation of protein kinase activity in rats with benign prostatic hyperplasia (BPH)and the mechanism of the inhibition of benign prostatic hyperplasia.Methods:Fortyfive SPF male Wistar rats were selected from our laboratory animal center.Rats were divided into three groups by random number table method.The observation group (15 rats),the model group (15 rats)and the normal group (15 rats)were selected.Rats in the model group and the observation group were used to prepare the BPH model.After successful modeling,the rats in the observation group were given intragastrically 1ml/100g of Qianlie Tongyu Decoction every day.The model group and the normal group were administrated with equal doses of normal saline for 30 days.After the last administration,the body weight,prostate wet weight,and prostate index were measured.The positive expression of PKC mRNA and PKA mRNA in the prostate tissue of rats was detected by in situ hybridization.The vascular endothelial growth in the prostate tissue of each group was detected by ELISA. Factors (VEGF),superoxide dismutase (SOD),epidermal growth factor (EGF),glutathione peroxidase (GPx)and malondialdehyde (MDA)content.Results:The prostate weight and prostate index of the model group were significantly higher than those of.the normal group.The prostate weight and prostate index of the observation group were significantly lower than those of the model group (P <0.05).The expression of PKC mRNA in the model group was positive.The rate of PKC/PKA was significantly higher than that of the normal group,and the positive expression rate of PKAmRNA was significantly lower than that of the normal group.The positive rate of PKCmRNA expression and PKC/PKA in the observation group Were significantly lower than those in the model group,and the positive expression rate of PKAmRNA was significantly higher than that in the modelgroup.All were statistically significant (P <0.05).The levels of EGF,VEGF and MDA in the observation gr
