摘要
目的:探讨人工栽培和野生银柴胡种质资源间ITS序列的差异性,为其种质资源的分子鉴定和品质评价提供参考。方法:ITS-PCR扩增产物双向测序后用DNAMAN软件处理测序数据,MEGA 5.0计算K-2-P遗传距离并用UPGMA法构建聚类图。结果:15份银柴胡种质资源共检测到10条不同的ITS序列,大小为625~626 bp,序列一致性达99.78%,遗传距离变幅范围0.002~0.013,所有序列均提交至NCBI并获登录号;10条序列中共发现12个SNP位点,UPGMA聚类结果显示野生银柴胡单独聚为一类。结论:所研究的人工栽培银柴胡和野生银柴胡种质资源ITS序列遗传差异小,亲缘关系近。
Objective:The differences of ITS sequences between artificial cultivation and wild SteUaria dichotoma var.lacceolata resources were discussed,which can provide reference for molecular identification and quality evaluation of Stellaria dichotoma vat.lacceolata germplasm resources.Methods :Stellaria dichotoma var.lacceolata sequencing data were processed by DNAMAN software after the bi-directional sequencing of ITS-PCR amplification products.The K-2-P genetic distance of SteUaria dichotoma var.lacceolata was calculated,and UPGMA tree was constructed by MEGA 5.0.Results:A total of ten ITS sequences which the fragment size ranged 625-626bp were detected in 15 varieties of Stellaria dichotoma vat.lacceolata germplasm resources,and the sequence similarity of these was 99.78%.The genetic distance ranged from 0.002 to 0.013.The sequences were submitted to the NCBI database and the accession number had been obtained.Twelve SNP locus were found in ten sequences,and wild Stellaria dichotoma var.lacceolata was clustered into a class by UPGMA clustering.Conclusion :The genetic diversity in ITS sequences of Stellaria dichotoma var.lacceolata is very small and has a close relationship between artificial cultivation and wild Stellaria dichotoma var.lacceolata.
