摘要
目的 :观察条件培养液对兔骨髓基质细胞向成骨细胞分化的影响。 方法 :分离兔骨髓 ,培养骨髓基质细胞 ,传代细胞分别用标准培养液和加入 β 甘油磷酸钠 ,地塞米松和抗坏血酸制成的条件培养液培养 ,观察不同培养液下骨髓基质细胞碱性磷酸酶活性和骨钙素含量。 结果 :在条件培养液下 ,骨髓基质细胞碱性磷酸酶活性和骨钙素含量明显高于标准培养液下的含量。 结论
Objectives:To study the osteoblastic differentiation ability of rabbit bone marrow stromal cells cultured in experimental culture medium. Methods:Bone marrow stromal cells isolated from rabbit bone marrow were cultured. Next, the passage cells were cultured in RMPI 1640 supplemented with 20% foetal bovine serum(standard medium) and in the presence of β glycerophosphate(β GP,10 mmol), dexamethasone(Dex 10 nmol) and ascorbic acid(AA,50 μg/ml). Cell morphology, ALP activity and ability to form osteocalcin(BGP) were observed and determined. Results:ALP activity and BGP were higher in the experimental medium. Conclusions: Osteoblastic differentiation/proliferation of rabbit bone marrow stromal cells were improved in the experimental culture medium.
