摘要
采用酸-酶结合法制备牛肌腱胶原纤维凝胶。研究了浓度、温度及pH值对胶原纤维凝胶黏度与吸水率的影响,用激光粒度仪测定胶原纤维凝胶的等电点,通过扫描电镜考察了胶原纤维凝胶海绵的断面、表面及胶原纤维束的形貌。结果表明:胶原纤维凝胶的等电点为4.22±0.04;胶原纤维凝胶经冷冻干燥后形成孔隙均匀的三维网状结构,孔径约为90-250μm,胶原纤维直径约为0.1~1.0μm。DSC分析测得胶原纤维凝胶海绵的热变性温度为66.8℃。在试验研究范围内,胶原纤维凝胶的黏度随其浓度的增高而增大,随温度的升高呈下降趋势,随剪切速率增大而降低。随pH值的升高,胶原纤维凝胶的黏度先降低后升高,胶原纤维的吸水倍率呈现升高后降低的趋势。当其pH值为3.5时,其吸水倍率达到289.8g/g,黏度降至最低值。
Collagen fibrous gels was prepared from bovine tendon by acid swelling-pepsin digestion method. The influence of the concentration, temperature and pH value of collagen fibrous gel on its viscosity and water-absorption rate were studied. It was found that the equipotential pH of collagen fibrous gels was 4. 22±0.04 by Zeta Poential Titrator, and the pore sizes of its sponge were about 90-250 μm and the diameters of collagen fiber were about 0. 1- 1.0 μm by SEM. The tests of Differential Scanning Calorimeter (DSC) revealed that the denaturation temperature of collagen fibrous gel sponges was 66.8 ℃. As the concentration decreased and the temperature and the shearing velocity increased, respectively, the viscosity of collagen fibrous gels was reduced. With the hoist of pH value, the viscosity of collagen fibrous gels was depressed first and upward at last, and its absorbent ability had an inverse tendency. While the pH value of collagen fibrous gels was 3.5, the absorbent ability of collagen fibers was 289.8 g/g for pure water and the viscosity dropped to the lowest.
