摘要 目的 :建立一种简单可靠的骨髓基质细胞 (BMSc)向成骨细胞转化的体外培养方法 ,探讨骨髓基质细胞向成骨细胞转化的机理。 方法:将兔 BMSc悬液进行体外培养 ,进行形态学观察和组织学检测。 结果:传代培养细胞的碱性磷酸酶 (AL P)阳性率达 80 %以上 ,表现为成骨细胞形态并形成钙结节。结论:地塞米松、β-甘油磷酸钠和维生素 C可促进 BMSc分化为成骨细胞 ,BMSc的分化和增殖是两个对立的状态。 Objective: To establish a simple and stabled method for the cultured bone marrow stromal cell (BMSc) in vitro to become osteogenetic cells. Methods: The BMSc were cultured in conditional medium in subculture including vitamin C,dexamethasone and β-glycerophosphate. The cells were examined by phase-contrast microscopy and histochemistry stains technique. Results: The cultured cells were spindle-shaped or polygonal-shaped. Histochemical studies showed positive stain of alkaline phosphatase activity in the osteoblast-like cells and within the mineralized nodules. Conclusions: Vitamin C,dexamethasone and β-glycerophosphate can induce BMSc to become osteogenetic cells. The proliferation and differentiation of BMSc are two opposed state that can not coexist.
