Pan-sequence analysis of Hantavirus Z37 strain genome

摘要 目的 测定Z37株的全基因组序列 ,了解我国汉坦病毒疫苗生产株分子基础 ,分析其遗传学和抗原性差异等。方法 设计出针对各片段的特异性引物 ,用PCR方法从Z37病毒感染的细胞提取细胞总RNA ,逆转录扩增、产物克隆T载体 ,纯化后测序 ,测定的序列应用DNASTAR软件比较分析。结果 Z37株全基因组由L片段 6 5 30nt、M36 5 1nt、S175 4nt组成 ,分别编码 2 15 1、1133、42 9个氨基酸 ,与同型病毒间同源率高达 95 .2 %~ 99.5 % ,绘出了 3个片段的核苷酸和氨基酸的系统发生树。结论 测定得到了Z37株的全基因组序列 ,为研究疫苗毒株的生物学特性。 Objective Analysis of Pan-sequence of the genome of Hantavirus Z37 strain which is applied for inactivated vaccine production in China for understanding its molecular characteristics and compare it with other Hantaviruses. Methods The total RNA was extracted from Z37 virus infected cells and the RT-PCR products were cloned into T vector, sequenced and analyzed by DNASTAR software. Results Z37 strain genome comprised of L 6530nt, M 3651nt and S 1754nt, encoding 2151, 1133 and 429 amino acids respectively. The homology of Z37 nucleotide sequences with that of other SEO type virus varied from 95.2% to 99.5%. Phylogenetic trees were constructed depending on nucleotide and amino acid sequences. Conclusion The genome sequence of Z37 strain might be used in studying its molecular mechanism, biological characteristic and pathogenesis.
