摘要 目的 :观察乙型肝炎病毒核心抗原 ( HBc Ag) DNA疫苗对小鼠和猕猴的免疫原性。方法 :肌内注射法接种 HBc AgDNA疫苗 ;EL ISA法检测小鼠和猕猴血清抗 - HBc Ig G终点滴度和 Ig G亚类水平 ;51铬释放法测定小鼠脾细胞 HBc Ag特异性细胞毒性T细胞 ( CTL )活性 ;EL ISA法检测猕猴外周血单个核细胞 ( PBMC)培养上清中 IFN -γ及 IL - 4水平 ;3H - Td R掺入法检测猕猴PBMC HBc Ag特异性增殖反应。结果 :小鼠和猕猴经接种该 DNA疫苗后均产生高滴度抗 - HBc抗体 (分别达 1∶ 3 2 80 5 0和1∶ 10 93 5 0 ) ;小鼠抗 - HBc Ig G亚类以 Ig G2 a为主 ,猕猴抗 - HBc Ig G亚类以 Ig G2占优势 ( Ig G1/Ig G2 <1) ;小鼠的 HBc Ag特异性CTL杀伤活性达 5 0 %以上 ;猕猴 PBMC培养上清中 ,IFN-γ水平与 IL - 4相比占明显优势 ( 15 .63 pg/ml vs <3 .13 pg/ml) ,猕猴PBMC抗原特异性增殖反应阳性 (刺激指数 =2 .12~ 3 .83 )。结论 :HBc Ag DNA疫苗对小鼠和猕猴均具有良好的体液免疫和细胞免疫原性。 Objective:To observe the immunogenecity of DNA vaccin e based on core gene of hepatitis B virus in both mice and rhesus monkeys. Methods:HBcAg DNA vaccine was intramuscularly immunized in this exper iment . Anti HBc antibody (IgG) and its isotypes in mice and monkey′s sera were dete cted by ELISA method. HBcAg specific cytotoxic T lymphocyte (CTL) activity in mi ce was measured by 51 Chromium release assay. IFN γ and IL 4 in the cult ure supernatant of monkey′s PBMC were measured also by ELISA. HBcAg specific pr oliferation activity of monkey′s PBMC was detected by 3H TdR incorporation a ssay. Results:High titers of serum anti HBc antibody were raised in b oth mice (1∶328 050) and rhesus monkeys (1∶109 350) after immunization with DN A vaccine of HBcAg. The levels of IgG2a and IgG2 isotypes were generally higher than IgG1 in mice and rhesus monkeys. HBcAg specific CTL activities were more th an 50% in both species of mice. In the culture supernatant of monkey′s PBMC IFN γ (15.63 pg/ml) was predominant, compared with IL 4 (<3.13 pg/ml). HBcAg spe cific proliferation activities of monkey′s PBMC were rather strong (SI=2.12~3.8 3). Conclusion:HBcAg DNA vaccine shows strong humoral and cellular i mmunogenecities in both mice and rhesus monkeys.
