摘要
目的 :U5 0 488H是阿片肽受体 κ亚型的选择性激动剂 ,本实验研究 U5 0 488H预处理是否可增强心肌细胞对热应激的耐受性 ,同时观察与热休克蛋白 70及 90表达的关系。方法 :采用分离的大鼠心肌细胞作为研究对象 ,以心肌细胞释放乳酸脱氢酶 (L DH)的多少反映其损伤程度并用 Western blot分析热休克蛋白表达的变化。结果 :在 5~ 30 μmol/ L 浓度范围内 ,U5 0 488H预处理 30 min剂量依赖性的减低心肌细胞热应激后 L DH的释放。同时热休克蛋白 70和 90的表达明显增加。结论 :U5 0 488H激活阿片受体后具有保护心肌细胞的作用 ,其保护机制可能与调节热休克蛋白的表达有关。
AIM: U50488H is the selective agonist of kappa opioid receptor. The present study was to investigate whether U50488H pretreatment increases rat ventricular myocytes tolerance against heat stress. METHODS: Ventricular myocytes were isolated and pretreated with U50488H for 30 min, then they were subjected to heat stress at 44℃ for 30 min followed by 2 h culture at normal conditions (37℃). RESULTS: The lactate dehydrogenase (LDH) release in the U50488H pretreatment group was decreased in the dose-dependent manner, and the expression of heat shock protein 70 and 90 were significantly increased. CONCLUSION: Kappa opioid receptor activation with U50488H increases rat ventricular myocytes tolerance against heat stress with elevation of levels of HSP70 and HSP90.
