摘要
目的 :探讨高纯度的蚯蚓提取物对人慢性粒细胞性白血病系K5 6 2细胞的作用及机制。方法 :以人K5 6 2细胞为靶细胞 ,采用细胞计数及台盼蓝拒染法、光镜观察、细胞凋亡荧光染色法、DNA凝胶电泳等技术 ,观察蚯蚓提取物对K5 6 2细胞生长的影响。结果 :① 10~ 2 0mg/L蚯蚓提取物可促进K5 6 2细胞的增殖 ;②≥ 5 0mg/L的蚯蚓提取物可通过凋亡轻度抑制细胞生长 ,表现为细胞呈凋亡形态学改变 ,DNA琼脂糖凝胶电泳出现梯形条带 ,细胞凋亡的荧光染色显示细胞凋亡的特征。结论 :小剂量的蚯蚓提取物可明显促进K5 6 2细胞的增殖 ,而高剂量则可诱导凋亡 。
Objective: To explore the effect and mechanism of the earthworm extracts on chronic myeloid leukemic cells. Methods: CML cell line K562 was used as target cells and cell count?trypan blue exclusion?light microscope examination?apoptosis fluorescence assay and DNA gel electrophoresis were assessed for the effect of earthworm abstracts. Results: ①At concentrations of 10-20mg/L could obviously facilitate the proliferation of K562 cell ②At concentrations of 50mg/L and above the cell numbers declined lightly. The morphology of apoptosis, characteristic DNA ladder on DNA gel electrophoresis appeared, and apoptotic cells were accounted for 30.3% by fluorescence assay. Conclusion: The earthworm extracts at low dose could facilitate the cells proliferation and on high concentration induced apoptosis. But the differentiation to mature cells was not seen.
