摘要
目的 探讨转化生长因子 (TGF - β1)对人视网膜色素上皮 ( (hRPE)细胞增殖的抑制作用。方法 用不同浓度的TGF - β1( 0 .0 1ng/ml、1ng/ml、10ng/ml)处理hRPE细胞 2 4h后 ,用MTT和氚 -标记脱氧核苷 ( 3H -TdR)方法检测TGF - β1对hRPE细胞和DNA合成的抑制率的影响 ,用流式细胞仪检测TGF - β1对hRPE细胞周期的影响。结果 不同浓度TGF - β1处理hRPE细胞 2 4h后细胞和DNA合成的抑制率随TGF - β1浓度的增加而增加 ,TGF - β1将细胞阻滞在G1期。结论 TGF - β1能抑制hRPE细胞的增殖且呈剂量依赖性 ,TGF - β1是通过将hRPE细胞阻止在G1期而发挥抑制作用的。
To investigate the inhibifing effect of transforming growth factor-β (1TGF-β 1) on the proliferation of human retinal pigment epithelial (hRPE) cells.Methods The cultured cells were treated with TGF-β 1 at varying concentrations of 0.01 ng/ml,0.1 ng/ml,1 ng/ml and 10 ng/ml for 24 h.Then,OD and cpm reflecting living cell number and cellular DNA synthesis were measured by MTT assay and tritium-labelled thymidine deoxyribose ( 3H-TdR) incorporation,respectively.Flow cytometry was used to analyse cell cycles.Results The inhibiting rates of cellular growth and DNA synthesis were directly proportional to the concentration of TGF-β 1 in hRPE cells;the percentages of cells in G 1 phase of cell cycle increased from 46.4% to 64.6%.Conclusions TGF-β 1 can inhibit the proliferation of hRPE cells dose-dependently at G 1 phase by .
