摘要
对人原代表皮细胞在无血清培养基 A、B和 C中的生长过程进行动态观察 ,并于培养第 5 d用 MTT法和3H- Td R掺入法分别测定其生长和代谢情况。结果表明 :人原代表皮细胞在无血清培养基 A中生长 (OD值 :0 .6 38± 0 .0 33)和代谢 (COM:92 8± 46 .6 9)优于无血清培养基 B(OD值 :0 .5± 0 .0 37,CPM:740 .11± 43.2 9) ,差异有极显著意义 (MTT法 :P<0 .0 1,3H - Td R掺入法 :P<0 .0 1)。在无血清培养基 B中人原代表皮细胞在培养第 12 d仍未汇合联接成片 ;而在无血清培养基 C中细胞的生长增殖与无血清培养基 A相似 (P>0 .0 5 ) ,细胞在这二种培养基中于培养第 12 d均可汇合联接成片。
The growth proceedings of human prokeratinocytes (HK) in serum free media (SFM) A,SFM B,SFM C were studied dynamically,and the growth and proliferation of HK was determined separately by MTT method and 3 H thymidine incorporation after 5 days′ culture.The results indicated:The growth (the value of OD:0.638±0.033) and proliferation (COM:928.00±46.69) of prokeratinocytes in SFM A was superior to that in SFM B (the value of OD:0.5±0.037,CPM:740.11±43.29),showing significant difference (MTT method: P <0.01, 3H thymidine incorporation: P <0.01).Human prokeratinocytes did not grow to be confluent monolayer cells after 12 days culture in SFM B.But after supplement of serum subsitute(Ss) in SFM B,the growth and proliferation of HK was similar to that in SFM A ( P >0.05).Human keratinocytes could grow to be confluent layered cells in SFM A and SFM C (SFM B+SS) 12 days after culture.
