摘要
目的 探讨神经生长因子 (nerve growth factor,NGF)对角膜上皮细胞增殖的影响 ,寻找促进角膜上皮损伤修复的有效方法。方法 在培养兔角膜上皮细胞的培养液中添加浓度为 5 u/ ml、5 0 u/ ml和 5 0 0 u/ m l的NGF,加药后的第 3、7天 ,采用四甲基偶氮唑盐 (MTT)法 ,于酶标仪上测定 5 70 nm波长处的吸光度值来观测细胞增殖情况。结果 浓度为 5 0 u/ m l和 5 0 0 u/ ml的 NGF对兔角膜上皮细胞增殖有明显促进作用 ,且两组间有差异 ,呈剂量依赖性 (P <0 .0 5 ) ,而浓度为 5 u/ ml的 NGF促细胞增殖作用不显著 (P >0 .0 5 )。结论 外源性 NGF对培养的兔角膜上皮细胞增殖有明显促进作用。表明 NGF具有临床应用的可能性。
Objective To study the effect of nerve growth factor(NGF) on proliferation of corneal epithelial cells.Methods Rabbit corneal epithelial cells in culture were treated with different concentrations of NGF(5u/ml,50u/ml and 500u/ml),On 3d and 7d after treated with NGF,MTT assay was used to examine the proliferation of the cells.Results 50u/ml and 500u/ml NGF enhanced cellular proliferation effectively and in a dose-dependent manner(P<0.05).The addition of 5u/ml NGF did not significantly enhance cellular proliferation(P>0 05) Conclusion The Results suggest that exogenous NGF can stimulate proliferation of rabbit corneal epithelial cells and it may be of potential use clinically.
