摘要 目的观察重组人表皮生长因子(recombinant human epidermal growth factor,rhEGF)对牛晶状体上皮细胞(lens epithelial cell,LEC)增殖的影响。方法体外培养牛LEC,在培养液中分别加入0μg.L-1、1μg.L-1、10μg.L-1、20μg.L-1、50μg.L-1、100μg.L-1、1000μg.L-1rhEGF,加药后72h采用MTT法,在酶标仪490nm波长处测定吸光度值并观测细胞增殖变化。结果0μg.L-1、1μg.L-1、10μg.L-1、20μg.L-1、50μg.L-1、100μg.L-1、1000μg.L-1的rhEGF作用于LEC72h后MTT法测得的OD值分别为0.166±0.029、0.195±0.017、0.232±0.034、0.263±0.019、0.315±0.009、0.261±0.005、0.206±0.043。结论rhEGF对牛LEC增殖有明显的促进作用,且呈浓度相关。 Objective To observe the effect of recombinant human epidermal growth factor (rhFGF) on proliferation of cultured bovine lens epithelial cell(LEC). Methods Bovine LEC were cultured in vitro and incubated with 0μg·L^-11μg·L^-110μg·L^-120μg·L^-150μg·L^-1100μg·L^-11000μg·L^-1hEGF for 72 hours. The effect of LEC proliferation was detected by methyl thiazolyl tetrazolium(MTT) after incubation. Results The OD value were 0.166±0.029、0.195±0.017、0.232±0.034、0.263±0.019、0.315±0.009、0.261±0.005、0.206±0.043 atμg·L^-11μg·L^-110μg·L^-120μg·L^-150μg·L^-1100μg·L^-11000μg·L^-1 concentrations of MTr,respectively. Conclusion The pro- liferation of LEC is promoted by rhFGF, showing dose-dependent.
