摘要
三裂叶葛无菌苗器官在MS + 1mg/LNAA + 1mg/L 2 ,4,5 T 培养基上培养 ,皆能形成愈伤组织 ,以子叶的诱导频率最高。B5+ 1mg/l2 ,4D + 0 .1mg/LNAA + 30 0mg/L水解酪蛋白用于愈伤组织继代培养 ,愈伤组织颗粒不断扩大 ,保持淡绿色。不同外植体形成的愈伤组织经继代培养产生异黄酮和葛根素的能力不同。
It was found that organs of sterile seedlings of Pueraria phaseoloides (Roxb.) Benth. formed Calli on MS medium contained 1 mg/L NAA and 1mg/L 2,4,5 T for 20 days, the callus induced frequency of cotyledon was higher than those frequency of leaves, stems, roots, embryonal axis and radicle . The callus growth increased on a subculture medium consisting B5 salts supplemented with 1mg/L 2,4 D, 0.1 mg/L NAA ,300mg/L casein hydrolysate and adjusted to pH 6.0. The production capacity of isoflavone and puerarin in calli formed from several organs were different.
