摘要
测定了不同的基本培养基,不同的激素水平,不同的糖含量以及不同的初始接入量对益母草悬浮培养细胞生长的影响.结果发现MS培养基最益于悬浮培养细胞的生长,3%的蔗糖水平细胞的生物量增长率最大.在初始接入量为6 g/L时细胞增殖最快,在低接入量的情况下细胞停止生长.在6-BA无论与2,4-D组合还是与NAA组合,均在细胞分裂素2 mg/L和生长素0.5 mg/L时悬浮培养细胞的增殖率达到最大.研究结果为细胞培养生产益母草生物碱提供参考依据.
Callus of Leonurus heterophyllus Sw. from solid medium were cultured in liquid medium, and the types of basal medium, concentration of plant growth regulators, content of sucrose in medium, and quantity of original incubated callus were respectively tested on the increase of callus proliferation. It was found that MS medium was most suitable for suspended cell growth, and the biomass of the cells increased rapidly in medium with 3% sucrose. 6g/L original incubated callus showed the most promptional effect on callus growth, however the cells stopped to growth ifvery small callus incubated in medium. In medium with combination of 6-BA (2 mg/L) and 2,4 -D (0.5 rog/L) or NAA consults for mass (0.5 mg/L) , suspended cells proliferated most rapidly. The results presented here provide the ire culture of L. heterophyllus and extraction and utilization of alkloid for this plant.
