生长素和细胞分裂素物质对野葛外植体器官发生的影响 被引量:13

EFFECTS OF AUXIN AND CYTOKININ SUBSTANCES ON ORGANOGENSIS OF PUERARIA LOBATA INVITRO

摘要 通过比较几种生长素和细胞分裂素物质对野葛(puerarialobata)外植体形态发生的影响发现,BA诱导的不定芽数量和质量均好于KT,最适浓度1mg·L1,同样浓度的BA比KT易使野葛外植体基部膨大和褐化,KT比BA更易使不定芽玻璃化,提高BA浓度或者不定芽在含BA或KT的培养基上继代,也会产生玻璃化和褐化,随继代次数增加而加重-在3种生长素类物质IAA、IBA和NAA诱导不定根形成过程中,在0-2mg·L-1以下,NAA诱导的效果最好,在0-3~0-4mg·L-1的浓度范围内,IBA效果最好,而大于0-5mg·L-1的浓度则IAA效果最好,在同样浓度条件下,NAA诱导的不定根粗短,而IBA和IAA诱导的不定根细长,多分枝,形态上较接近自然根-3种生长素类物质诱导愈伤组织和褐化能力依次为NAA>IBA>IAA,低浓度下生长素物质诱导的不定根生长快、生长质量好,幼苗移栽成活率高,而过高的生长素物质诱导的根系粗短,易褐化和形成愈伤组织、幼苗移栽不易成活,反映不定根与外植体维管束联系差-当BA与IBA混合使用时,BA会严重抑制IBA诱导的不定根形成,而IBA对BA促进的不定芽分化效应作用不明显。 By comparing the effects of some auxin and cytokinin substances on organogensis of explants of Pueraria Lobato, it was found that the number and quality of adventitious buds induced by BA were better than those induced by KT, with the optimum concentration being 1 mg/L. The same concentration of BA caused swelling and browning of Pueraria Lobata explant basal more easily when compared to KT, but KT caused hyperhydriciting of adventitious buds more easily than BA. Increasing BA concentration or repeatedly subculturing adventitious buds in medium containing BA or KT also caused hyperhydriciting and browning. As far as the formation of adventitious roots induced by IAA, IBA and NAA was concerned, NAA yielded the best results below the concentration of 0.2 mg/L, IBA showed the best effects during 0.3~0.4 mg/L, while above 0.5 mg/L, IAA was the best. Under the same concentration, roots induced by NAA were thicker and shorter while roots induced by IAA and IBA were smaller and longer and had more branches. The capacity of inducing callus and browning followed the order of NAA>IBA>IAA. Roots induced by low concentrations of auxins grew well and had higher survival rate of plantlet transplanting. On the contrary, roots induced by high concentrations of auyins were thicker and shorter, browned and formed callus more easily, with a low survival rate of plantlet transplanting, indicating the poor interaction between buds and roots. When BA and IBA were mixed, BA seriously inhibited the formation of adventitious roots induced IBA, while no obvious effects of IBA on adventitious buds induced by BA were achieved. The optimum prescription for producing adventitious buds and roots at one step was BA 0.5 mg/L+IBA 0.3 mg/L.
