Effect of NGF and passive movement on bone metabolism in rats with denervation

摘要 目的拟观察外源性神经生长因子对去神经大鼠血清BGP和TRACP的影响,探讨神经生长对废用性骨质疏松的预防作用。方法 50只大鼠随机抽签法分为5组:假手术组(SHAM)、去神经组(DN)、神经生长因子组(NGF)、被动运动组(PM)、神经生长因子与被动运动联合组(NGF+PM)。假手术组不切除坐骨神经和股神经,其他各组通过切除大鼠右侧坐骨神经和股神经建立SD大鼠右侧后肢的去神经模型,通过被动运动和外源性生长因子的干预措施,观察各组大鼠血清BGP和TRACP指标。结果与假手术组相比,去神经各组血清BGP方面明显降低(P<0.01),血清TRACP明显升高(P<0.05);与DN组相比,NGF组和NGF+PM组与之都有显著性差异(P<0.05),PM与DN组之间虽无统计学意义(P>0.05);与DN组血清TRACP相比,PM组和PM+NGF组与之有显著性差异(P<0.05),NGF组与之无差异(P>0.05);结论大鼠下肢缺乏神经支配导致肌肉萎缩、骨正常代谢遭到破坏,被动运动和神经生长因子能提高去神经大鼠血清BGP水平、降低血清TRACP的含量,一定程度上能预防去神经型废用性骨质疏松的发生。 Objective To investigate the effect of exogenous NGF on the serum BGP and TRACP in denervated rats, and to explore the preventive effect of nerve growth on disuse osteoporosis.Methods Fifty rats were randomly divided into 5 groups:sham operation group ( SHAM) , denervation group ( DN) , NGF group ( NGF) , passive movement group ( PM) , and combination group ( NGF+PM) .Rats in SHAM group were injected with saline, without the excision of the sciatic nerve and the femoral nerve.Rats in other 4 groups were all induced disuse osteoporosis by the excision of the sciatic nerve and the femoral nerve.The serum levels of BGP and TRACP were observed.Results Compared with that in SHAM group, BGP level in the other 4 groups decreased significantly (P〈0.01), but TRACP level increased significantly (P〈0.05).Compared with that in DN group, BGP level in NGF group and NGF+PM group was significantly different (P〈0.05), while no significant difference between PM group and DN group was observed ( P〉0.05 ) .The serum TRACP level in PM group and PM+NGF group was significantly different with that in DN group (P〈0.05), while no significant difference between NGF group and DN group was observed (P〉0.05). Conclusion Denervation in rat lower limbs leads to muscular atrophy and destroyed bone metabolism .Passive movement and nerve growth factor can improve the serum BGP level and reduce the serum TRACP level in denervated rats , showing a preventive effect on the disuse osteoporosis to a certain extent .
