不同浓度益气活血方含药血清对大鼠骨髓间充质干细胞体外增殖及VEGF表达的影响 被引量:4

Effects of Yiqi Huoxue Formula(益气活血方) Contained Serum of Different Concentrations on the Proliferation of BMMSCs and the Expression of VEGF in Rats

摘要 目的探讨不同浓度益气活血方含药血清对大鼠骨髓间充质干细胞(BMSCs)增殖活力及血管内皮生长因子(VEGF)的影响。方法将8只SD大鼠随机分为空白对照组4只、益气活血方组4只,纯化培养BMSCs,采集空白对照组(灌胃蒸馏水)、益气活血方组(灌胃益气活血方)大鼠血清;将BMSCs分为空白血清对照组、5%、10%、20%益气活血方血清组,分别以含10%空白血清及含5%、10%、20%益气活血方血清培养液干预培养BMSCs。MTT法检测BMSCs增殖活力,ELISA法检测培养BMSCs上清液VEGF水平。结果第2天、第4天、第6天,10%益气活血方血清组BMSCs去基线增殖率较5%益气活血方血清组与20%益气活血方血清组明显增加(P<0.05或P<0.01);第2天、第4天组间BMSCs上清VEGF水平比较,10%益气活血方血清组明显高于空白血清对照组、5%益气活血方血清组、20%益气活血方血清组(P<0.01)。结论 10%益气活血方含药血清可明显提高BMSCs增殖能力,促进VEGF表达。 ( 1. China Academy of Chinese Medical Sciences) ABSTRACT Objective To research the effects of Yiqi Huoxue Formula (qi-supplementing blood-quickening formula, 益气活血方) contained serum with different concentrations on the proliferation of bone marrow mesenchymal stem cells (BMMSCs) and the expression of vascular endothelial growth factor (VEGF) in rats. Methods Eight SD rats were randomized into blank control group and Yiqi Huoxue Formula group with 4 rats in each group. The BMMSCs were purified and cultured in rats. The serums were collected in the blank control group ( intragastric admin- istration with distilled water) and Yiqi Huoxue Formula group (intragastric administration with Yiqi Huoxue Formula). The BMMSCs were divided into the blank serum control group (BMMSCs were cultured by 10% blank se- rum) and 5% , 10% and 20% Yiqi Huoxue Formula serum groups (BMMSCs were cultured by 5% , 10% and 20% Yiqi Huoxue Formula serums respectively). The proliferation activity of BMMSCs was measured by MTT assay. The expression of VEGF in cultured BMMSCs supernatant was detected by ELISA assay. Results Comparing with the 5% and 20% Yiqi Huoxue Formula serum groups, the baseline removal proliferation rate of BMMSCs in the 10% Yiqi Huoxue Formula serum group was significantly increased on the 2nd, 4th and 6th day (P 〈0. 05 or P 〈0. 01 ). Comparing with the blank serum control group and 5% and 20% Yiqi Huoxue Formula serum groups, the levels of VEGF in the 10% Yiqi Huoxue Formula serum group were significantly increased on the 2nd and 4th day (P 〈 0. 01 ). Conclusion Yiqi Huoxue formula contained serum at 10 percent can significantly improve the proliferation of BMMSCs and promote the expression of VEGF.
